Rad1 was identified in yeast as a component of the checkpoint complex using checkpoint mutants. The human homolog of Rad1 has been shown to form a stable radioresponsive checkpoint complex with the human Rad9 and Hus1 proteins. This 32 kDa complex is present in undamaged cells but undergoes extensive modification in response to DNA damage. Human Rad1 exhibits homology with the Ustilago maydis Rec1 exonuclease and may possess a 3’- 5’ exonuclease activity.
